Therapeutic window: when to intervene before senescent load is irreversible — the object is not a ceiling that a senolysis calendar can date
1. Executive scientific abstract
The verified evidence does not support the chain “there is an ovarian senescent-load threshold; intervening before it reverses the state; afterwards it is already irreversible; that extends women’s health or life.” The title fuses four objects that do not substitute for one another: a therapeutic (none of L10-2 or L10-3 is identified as ovarian senolysis), a clock, a load (absolute number of a qualified durable arrest) and an irreversibility (of what: death, niche, endowment or function). After labelling each inherited primary with a single clock, no t1/t2 pair of the natural estimand remains at matched occupancy. García 2024 already started D+Q or fisetin from ~1 month and from 6 months; follicles, pregnancy and litter do not separate in either, and the regimens are not the same occupancy (three days every two weeks versus three days per month).
The only time series that publishes both a fraction called senescent and primordial counts —Yan et al. 2024, PMID 38871781, PMC11176158— does not fill the ceiling. The abstract says senescent primary oocytes “accumulated in aged ovaries.” The body writes continuous loss: primordials 1687 ± 498 (2 months) → 284 ± 72 (9 months) → 47 ± 14 (18 months); translocated HMGB1 7.97 ± 0.18% → 15.6 ± 1.41% → 26.8%; n = 5; mean ± SD. The inferred product of means falls ~134 → ~44 → ~13. The methods already define the whole-ovary integer (senescent oocytes/section × total sections, every fifth 10 µm section) and the results do not publish it as a series. Lamin B1, IL-1α, IL-6 and SA-β-gal are scored on 12 middle sections: they are not that integer. p21 is ubiquitous; p16-3MR is luciferase+ in all primaries; TUNEL and caspase 3 of that object are null at 2 and 9 months. The germ cell was already in prophase I. That does not pass Q0-STATE.
ABT-263 (navitoclax) 50 mg/kg/d, 7 on / 21 off × 3 months from 3 months, is a systemic t1 of another drug. It lowers the HMGB1 fraction and raises primordials (772 ± 38 vs 951 ± 46 at 6 months; 244 ± 27 vs 343 ± 56 at 9 months, n = 5). Senolysis of a ceiling does not predict more endowment. That sign is not the Flaws 2001 census: the perinatal surplus is already gone by P30–60, and Yan gavages adults. PBE 18.8% vs 43.1% is in-vitro maturation, not litter. The litter 5 ± 5 vs 8 ± 7 remains an internal conflict and no sign is chosen. D+Q and fisetin remain H0. The alkylator t_pre that does move structure is PTEN–PI3K–FOXO3 (Castrillon, Reddy, Li), not a SASP threshold. Sato 2026 puts the p16 maximum in corpus luteum and albicans (5.1× and 7.3×); ELISA without total protein. Hense 2024 is a review: it glosses “prevent, cannot reverse” as endowment, not as an experiment of qualified senescence.
The adversarial review split the generation primary. L10-4-HG-H1 required five clauses at once and called Flaws census the adult ABT; nested IL+ is not another cell (all IL+ ⊂ translocated HMGB1 of the same dictyate). L10-4-HG-HT1 used AMH as primordial census; Weenen 2004 does not observe AMH in human primordials. The operational hypotheses of this report are L10-4-AR-H1 v1, L10-4-AR-H2 v1 and L10-4-AR-HT1 v1. Computation was skipped: GSE253194 is bulk n = 3 and is not D0-ABS-EXTRACT.
Scientific delta of this report (L10-4-REPORT-EN-C1): the English twin freezes the same bilingual factual base and corrects the recorded bilingual-QA error: a Spanish synonym of wording matched a forbidden legacy term; both twins now use wording/redacción. The hypotheses that this twin must carry are those of the adversarial review, not the five-clause conjunction nor SCOPE-H1 on dictyate. D0-ABS-EXTRACT is restricted to the whole-ovary HMGB1 integer that Yan already computed in methods and does not publish; lamin/IL/SA-β-gal remain a fraction of 12 sections. If that series is not estimable, the recount is a physical disector of the same series at 2 versus 9 months, n = 7 per age (power ≈ 0.82 for a +50% rise if CV ≈ 0.30), not a contralateral class × stain product and not a third time point at 18 months on 47 ± 14 primordials. AMH does not enter as census. Flaws P0–P60 is not rerun as the adult ABT clock. G8 stays closed.
The operational conclusion is negative and actionable. There is no ovarian object whose absolute count of a durable arrest, female as n, rises to a written ceiling that splits function at matched occupancy. Dating “when to intervene” without that object is an empty protocol. The smallest next step is not another flavonol gavage nor an AMH RCT: it is to read or recount the integer Yan already defined.
2. Scientific question and relevance
The question is which mechanisms connect a therapeutic window —intervening before senescent load is irreversible— to women’s health and longevity, and which new hypotheses can be falsified. Answering it requires separating objects that the title fuses:
- What is the therapeutic whose calendar is being dated? D+Q, fisetin, quercetin alone, navitoclax, genetic clearance of a qualified lineage, or none.
- Is the “when” chronological age, STRAW+10, female months, week relative to a gonadotoxic, pulse versus daily, or a load/structure threshold written before seeing function?
- What accumulates: absolute number of a durable arrest, captured fraction, lipofuscin, infiltrate, or composition (Isola: immune and CD90+ at 9 months)?
- What ceases to be reversible: selective death, niche/fibrosis, the primordial pool, or an independent function?
- Does load precede loss of function, or does depletion produce the marker that is called load?
- Does “early vs late” live in young injury, in D-gal, in VCD, or in the uninjured adult female?
- Is a window trial in women missing, or are links 1–6 missing?
The relevance is high because “intervene in time” is a programme slogan and a cheap category error. The ovary is especially vulnerable: p16, p21, SA-β-gal, cytoplasmic HMGB1, lipofuscin, CD68, AMH, follicles and litter are not interchangeable. Neither are prevention one week before cisplatin, Gao after doxorubicin, and García’s two natural starts. A longer reproductive life can associate with skeletal or metabolic benefit and, at the same time, with greater exposure to hormone-dependent risks (L10-1-K10). The correct unit is an organ event with mediation and competing risks, not a reproductive surrogate.
3. Scope, population and life stage
The primary biological estimand is the adult female in natural reproductive aging, without a gonadotoxic. The contrast is the same intervention at the same exposure, applied at two biological times defined by a load or structure threshold written before the phenotype, on two coprimaries: (i) selective death of a state that already passed Q0-STATE; (ii) an independent ovarian function. Female months, STRAW and milligrammage are not that threshold. The inferential unit is the female or the woman. Follicles, sections and wells are not replicates.
For human translation, the relevant population is women classified by STRAW+10: late reproductive (−3b/−3a), transition (−2/−1) and early postmenopause (+1c), analyzed separately. Chronological age alone does not substitute for stage. Jin’s young/old atlas contrast (23–29 vs 49–54, without STRAW; illegitimate ovary47) is not this population.
The doxorubicin (Gao), D-galactose (Dong/Zhu/Niu), VCD (Ávila, abstract) and cisplatin/cyclophosphamide t_pre (Du, Xu, Su, Li) models are retained as toxic-calendar contrasts. They inform cytoprotection, marker–structure dissociation or a null of induced marker; they are not averaged with the natural transition. PCOS, ischemia–reperfusion, ovarian cancer and male evidence transported without a female replicate remain outside. Farr, Hickson, Justice, ITP, Krishnakumar and Baker are extraovarian: they do not fill G7.
Prescription, dose recommendation, senolytic ranking, synergy with NMN (10.5), an observational protocol (10.6), user data, identification of García’s Cdkn1/Serpin1/Ccxl5, SERPINE1 as an analyte, p>0.05 as irreversibility or equivalence, and any inference from fertility or menopause to longevity without an organ event are excluded. CD38 remains parked. G8 stays closed.
4. Background and mechanistic map
4.1 Operational definitions
- Therapeutic window: a biological-time interval in which a local intervention changes an independent function, and outside which the same intervention, at the same exposure, does not. It is not a calendar month. It is not a STRAW stage until anchored to a load or structure threshold.
- Senescent load: absolute number of cells of an identified lineage that meet durable arrest plus an orthogonal readout, failed in luteum, atresia and cycling cells of the same female or donor. Atlas fraction,
CDKN1A+,CDKN2A+, SenMayo, lipofuscin, CD68, IL-6, cytoplasmic dictyate HMGB1 andSERPINE1are not load. - Irreversible: one of three readings, declared before the datum: (a) the qualified cells no longer die at the exposure that used to kill them; (b) they die and fibrosis or endowment do not return; (c) they die and independent function remains non-separated. A
p>0.05null is not irreversibility. Q0-STATE: acquired durable arrest plus orthogonal, failing in luteum, atresia and cycling cells of the same female. Constitutive primordial prophase I is not that arrest (L10-4-MS-R1).- Ovarian function: a readout independent of the marker. AMH, estrous regularity and litter are not that function and are not longevity.
- Healthspan/longevity: an organ event, mediation and competing risks. Remains H0.
4.2 Favored chain that the literature would have to demonstrate
Lineage that already passed Q0-STATE
→ absolute number (not fraction) that rises to a threshold written BEFORE function
→ same intervention, same free species, t1 below the threshold and t2 above it
→ selective death of the qualified state versus sisters
→ independent ovarian function splits
→ (niche branch) fibrosis/PMF do not return even if death continues
→ systemic trajectory
→ organ event in women, with competing risks
Current literature breaks at the first link. There is no qualified state on the primordial. There is no natural t1/t2 pair at matched occupancy. There is no identified senolysis. The bridge to an organ was not tested.
4.3 Competing mechanisms
A — there is no therapeutic to date (H2). García: two calendars, quiet function. D+Q and fisetin H0.
B — dictyate category error (MS-R1). Yan scores a germ cell already in prophase I with an alarmin (cytoplasmic HMGB1) that is not Davalos’ (nuclear→extracellular, TLR4→IL-6 in a mitotic fibroblast).
C — the point of no return is endowment (H5). Finite pool. Fraction↑ × pool↓ = parental absolute↓. BCL2 sets the perinatal census (Ratts/Flaws/Morita) and Gao moves it up with fisetin.
D — toxic or regimen clock (H3). Alkylator t_pre via FOXO3 vs Gao post-doxorubicin vs Dong daily vs García pulse. L10-3-K8.
E — marker down, structure quiet (H4). Gao; García lipofuscin.
F — luteum/p16/p21 (H6). Sato CL/CA; Meinsohn reversible; Jirawatnotai; L7-4.
G — load or niche threshold (SCOPE-H1). No ovarian link. Relocated to a mitotic Q0 or to natural fibrosis (AR-H2 2b/2c), not to nested IL+ nor to cycling epithelium.
5. Evidence method
The complete artifacts of scoping, map, verification, synthesis, computational decision, hypotheses, adversarial review and experimental design were integrated. Claims were kept by layer: human (H), animal (A), in vitro/ex vivo (V), extraovarian (X), computational (C) and inferred (I).
D0-CLOCK-MAP labelled each primary with a single dominant clock (toxic, regimen, female calendar, STRAW, composition, absolute load, extraovarian). None pass Q0-STATE. After that label, G0-CLOCK closes.
Yan 2024, Sato 2026, Davalos 2013, Flaws 2001, Weenen 2004, John 2008 and Myers 2004 were reread in body or audited abstract in the stages that sign them. Gao, García, Du/Xu/Su, Dong, Fang, Isola, Farr and ITP are inherited as verified from L10-1/L10-2/L10-3. Hense 2024 is read as a review, not as an experiment. Watson remains a preprint. Ávila remains at abstract; n and dose are not invented. Farr: n = 60, CTx P = 0.611; the map’s 62–88 range is not retained (absent from the audited abstract). ITP: fisetin from 20 months, not 12.
p>0.05 was never interpreted as equivalence nor as irreversibility. Sections, fields and wells were not promoted to female number. Yan’s 15% was not treated as absolute. The product of means ~134 → ~44 → ~13 is labelled inferred, not stereology. section × sections counts are not a fractionator. The litter 5 ± 5 vs 8 ± 7 is not resolved by choosing wording or figures.
No computation was run (decision: skip). GSE253194 is bulk n = 3 and does not contain raw oocyte n. BioNeMo does not measure translocation.

7. Contradictory evidence and null findings
- Yan abstract vs body. “Accumulated in aged ovaries” is fraction accumulation over a collapsing pool. The body: “senescent primary oocytes may be constantly lost.” Title 10.4 inherits the abstract’s wording (
L10-4-HG-C1). - Yan orthogonals do not vote the same absolute. Inferred HMGB1 and lamin B1 fall; SA-β-gal ~flat (~30 → ~25); IL-1α/IL-6 rise by a digit (~10 → ~17 and ~7 → ~14) over events 0.44–5.9%, n = 5. Three readings of “load,” three signs (
L10-4-VER-R1). - Two natural calendars, quiet function. García exp. 1 and exp. 2. Lipofuscin can fall. That kills “intervening before 6 months is enough” (
L10-4-SCOPE-C3). - t_pre vs t_post. Du/Xu/Su move structure; Gao does not restore it. Averaging them fabricates the window (
L10-4-EV-C3). - Regimen. Dong daily ≠ García monthly ≠ Fang 1 dose/month ≠ ITP diet 600 ppm. This is not load irreversibility (
L10-3-K8). - ABT sign vs ceiling. More primordials when the fraction falls. Incompatible with “I cleared a senescent stock.” Not the Flaws census (
L10-4-AR-C1). - Yan litter. Wording “more pups” vs 5 ± 5 (ABT) and 8 ± 7 (control). No sign. PBE IVM does not substitute.
- p16 does not identify Yan’s object. p16-3MR luciferase+ in all primaries. Sato puts the maximum in CL/CA.
- Late-life extraovarian still operates. Baker 2011/2016. The female ITP null at 20 months does not prove that García’s ovary had already crossed a threshold.
- Human. Zero ovary under a senolytic. Farr: primary CTx null. Watson: preprint, not G7.
- Nested is not another cell. All IL+ had translocated HMGB1. Relocating
SCOPE-H1there does not escapeMS-R1(L10-4-AR-C3). - AMH is not the primordial. Weenen 2004. A model that “beats AMH” may be beating active folliculogenesis (
L10-4-AR-C4).
8. Multiscale mechanistic synthesis
8.1 Protein: three nodes, three clocks
HMGB1. Davalos: in a mitotic senescent fibroblast, nuclear HMGB1 relocates to the extracellular medium; redistribution requires p53, not ATM; secreted oxidized HMGB1 drives IL-6 via TLR4 (L10-4-MS-C1, V/X). Yan scores cytoplasmic translocation (or nuc−/cyto−) in an MSY2+ oocyte already in prophase I, apoptosis-negative, with nested intracellular IL-1α/IL-6. Extracellular and neighbour TLR4 were not measured. Importing the alarmin does not qualify Q0.
BCL2 / BCL2L1 / BCL2L2. ABT-263 inhibits the triad to “clear.” Ratts: bcl-2−/− is born with fewer primordials. Flaws: OE increases endowment at birth; by P30–60 the surplus is gone. Morita: Bcl-2 under ZP3 in the growing oocyte, not primordial; atresia↓, litter quiet. Gao: Bcl2 mRNA↑ with fisetin, TUNEL null. The lever is dual-use. An ovarian BH3 does not split H1-cell from H5 (L10-4-MS-C2). The adult sign 951 vs 772 is not that perinatal census (L10-4-AR-C1).
PTEN–PI3K–FOXO3. Castrillon: Foxo3a−/− activates the pool → secondary infertility. Reddy: oocyte-specific Pten activates the entire primordial pool → POF. Li: quercetin before CTX brakes the same axis. That is the t_pre molecule, not a SASP threshold (L10-4-MS-C3). John 2008 leaves shuttling (nuclear / cytoplasm / mixed) as the adult dormancy assay, FVB/n background.
8.2 Cell: an arrest is not piled on an arrest
The primordial default is arrested meiosis. Q0 asks for an acquired arrest that fails in luteum, atresia and cycle. p21 is in all types. p16-3MR is in all primaries. TUNEL/caspase 3 null at 2 and 9 months. The germ cell fails Q0 by construction (L10-4-MS-R1). Nested IL+ lives inside that parental class. The human epithelium that rises in fraction is cycling (L10-4-HG-C3): it is not the lineage to which to relocate H1. The rational residue is a non-luteal CDKN2A+ minority (~0.43% of nuclei, L10-1), with Q0-R2-IDENTITY unrun.
8.3 Tissue: four irreversibilities, one title
- Selective death. No ovarian instance of a Q0 lineage that dies at t1 and stops dying at t2.
- Niche / fibrosis. Gao: Il6/Mcp1↓, fibrosis/γH2AX quiet (injury). Isola: collagen↑ MMP2↓. CellChat downward: the opposite sign of a propagating SASP field. Watson preprint does not fill G7.
- Endowment / composition. Non-renewable pool. Yan: fraction↑ absolute↓. Gao: D+Q alone reduces PMF. Isola: immune/CD90+.
- Luteal identity / STRAW. Sato: ovulation 42.3 ± 3.1 → 15.6 ± 1.9; fertilization conserved; implantation 78.4% → 38.1%. ELISA quarantined. L7-4: luteal exposure falls without identified senescence.
8.4 System: the ovary of women is not in the chain
Baker late-life extraovarian still operates and does not name ovary. ITP fisetin from 20 months, female median null, no ovary. Farr CTx P = 0.611, n = 60, no ovary. Krishnakumar: 3 women, plasma aglycone, no ovary. G7 remains at the first link: qualified ovarian identity. Late ANM splits bone/T2D and hormone-dependent cancer. G8 closed.
Title 10.4 fuses
senescent ceiling ── dictyate + mislocalized HMGB1 + parental absolute↓
finite census ── perinatal BCL2 ≠ adult ABT; pool does not renew
toxic t_pre ── FOXO3/PI3K, not SASP
PK regimen ── pulse vs daily (L10-3)
luteal p16 ── Sato CL/CA
extraovarian “late” ── Baker still operates; ITP/Farr do not vote ovary
9. Computational layer
decision: skip. GSE253194 is bulk whole-ovary RNA-seq, 24 samples, n = 3; ages 3/6/9/12/18/25 months plus ABT-263 only at 6 and 9; deposit GSE253194_readcount.txt.gz (1 028 575 bytes, 54 532 Ensembl genes, FTP 2026-09-29). That is not D0-ABS-EXTRACT. Bulk Il6/Hmgb1/Tlr4/Bcl2 does not distinguish dictyate plus endowment from a SASP+ ceiling: RNA is not translocation nor a qualified absolute number, and Isola already doubled immune at 9 months. The RNA clock is not the n = 5 histology clock. NCBI GEO, 2026-09-29: ovarian navitoclax = only this series. BioNeMo does not enter: there is no variant or fold that measures translocation, stereology or a t1/t2 pair. No matrix was uploaded to an external service (L10-4-CD-C1–C3).
10. Primary hypothesis
L10-4-AR-H1 v1 — Yan’s primordial is not a ceiling; adult ABT is not the Flaws census; clauses are voted separately
Role: operational primary. Splits L10-4-HG-H1 v1 (conjunction withdrawn; census identity withdrawn). Absorbs MS-R1, HG-C1, VER-C1–C2, AR-C1–C3. Does not confirm SCOPE-H1. Does not rank D+Q against fisetin.
Falsifiable statement: in the uninjured adult female, the absolute number per female of HMGB1-translocated primary oocytes —the whole-ovary integer Yan already defines— and of lamin B1− from 12 sections, labelled as a spatial crop, do not rise to a threshold written before an independent ovarian function; p21, p16-3MR and prophase I do not qualify Q0 in that lineage; nested IL-1α/IL-6 from 12 sections, if it replicates, is interpreted first as enrichment or noise of the same parental class, not as another lineage. The ABT sign of more primordials at 6–9 months is treated as incompatibility with “cleared ceiling,” not as the Flaws 2001 census. D+Q/fisetin and FOXO3 t_pre are recorded as named clauses, not as coprimaries of this statement.
Lineage: L10-4-HG-H1 v1 (weakened); L10-4-MS-R1; L10-4-HG-C1; L10-4-VER-C1; L10-4-VER-C2; L10-4-AR-C1; L10-4-AR-C2; L10-4-AR-C3; L10-4-ED-C2. Yan PMID 38871781. Flaws PMID 11259262. Davalos PMID 23649808.
Predictions:
D0-ABS-EXTRACT: whole-ovary HMGB1 integers per female do not cross a threshold written before function. Nested 12-section objects are not relabelled mitotic nor whole-ovary integer.- If EXTRACT is not estimable,
D0-ABS-RECOUNTat 2 vs 9 months, n = 7, same series: the mean HMGB1 integer at 9 months does not exceed that at 2 months, or the +50% rise remains non-estimable. A 9 vs 2 with p > 0.05 is not read as “the parental does not rise.” - Stereology or raw n preserve the downward direction or the noise. If the parental rises and splits function at matched PMF, this primary dies at (i). Q0 remains failed: that does not reopen
SCOPE-H1on dictyate. - Matching García occupancy is not interpreted as a t2 of this object.
- GSE253194 does not vote.
Evidence for (A): Yan, written continuous loss; inferred product downward; discordant orthogonals; Q0 failed; García quiet function. (V/X): Davalos ≠ stain. (A): Flaws P30–60 ≠ Yan 3 months.
Evidence against: single-digit nested IL+ (VER-R1); n = 5; no stereology; apoptosis of ABT clearance not shown; PBE IVM moves; nobody applied Q0 to a mitotic (AR-H2).
Kill: mitotic Q0 absolutes (not dictyate) rise and split function at matched PMF and occupancy; or Q0 passes in the primordial as an acquired arrest separable from prophase I.
Status: proposed_primary.
Maturity: H1 of literature for dictyate + downward parental + failed Q0. H0 for “no ovarian lineage will have a ceiling” and for formal García equivalence.
Confidence: 0.70 that Yan’s primordial is not a senescent stock that crosses a ceiling; 0.12 that adult ABT is the Flaws census (withdrawn); 0.28 that no qualified mitotic will do so.
11. Competing hypothesis
L10-4-AR-H2 v1 — three rivals, not a nested disguised as another cell
Role: competing. Splits L10-4-HG-H2 v1. Nested IL+ returns to AR-H1 as a parental clause.
Falsifiable statement: after excluding the primordial oocyte in prophase I as a ceiling object, one of these three —declared before the datum— explains the residue: (2a) the primordial rise under ABT at 6–9 months is dormancy/counting/systemic (nuclear FOXO3 or growing:primordial ratio; or the disector dissolves 951 vs 772); (2b) a mitotic lineage of the same female passes Q0 (durable arrest + orthogonal, failed in luteum, atresia and cycling epithelium, without post hoc exclusions) and its absolute rises to a threshold written before function, at matched occupancy; (2c) a fibrosis/PMF threshold in natural aging splits function below and not above (H1-niche), distinct from Gao post-doxorubicin. Watson preprint does not fill (2c). Sato CL/CA is not (2b).
Lineage: L10-4-HG-H2 v1 (weakened); L10-4-VER-R1 (returned to parental); L10-4-HG-C3; L10-4-ED-C4; L10-4-ED-C5. Castrillon PMID 12855809. Reddy PMID 18239123. John PMID 18601916. Gao PMID 36648735.
Kill of (2a): fractionator + pinned FOXO3 conserve pool↑ and there is neighbouring Q0 death.
Kill of (2b): Q0 fails in the non-luteal CDKN2A+ minority of two replicates (2 and 9 months), without new exclusions; floor ≥20 non-luteal p16 nuclei/female.
Kill of (2c): fibrosis moves with the primordial census and not with a qualified state, or appears only post-toxic.
Status: proposed_competing.
Maturity: H0–H1. H1 of literature for marker–fibrosis dissociation in injury and for FOXO3 of dormancy. H0 for a natural threshold and for nested-as-another-cell.
Confidence: 0.44 in (2a) as a reading of the ABT sign; 0.22 in (2b); 0.18 in natural (2c); 0.08 in nested-as-lineage (withdrawn).
12. Translational hypothesis
L10-4-AR-HT1 v1 — the rule of not substituting the organ includes not calling AMH a census
Role: translational. Splits L10-4-HG-HT1 v1. HUMAN_QA_REQUIRED. Not a biomarker. Not a product. Not 10.6. Not prescribed.
Falsifiable statement (design rule, already in force): in adult women classified by STRAW+10 (−3b/−3a, −2/−1, +1c separately), outside any user cohort, a parameter made of AMH, antral count, menstrual regularity, age at menopause, T-lymphocyte p16, Farr CTx/P1NP, ITP median, Baker adipose/kidney/heart, or a module called ovarian senescent load without Q0 identity will not be accepted as mediator of an organ event. AMH, by Weenen 2004, is labelled growing pool if it ever enters as a covariate, not as primordial endowment.
Empirical form (parked): none of those substitutes is run until AR-H1 or AR-H2(2b/2c) deliver an object. The comparator, if that object exists, is STRAW stage + a primordial or histological census named as such + systemic inflammation, with competing risks. Beating AMH does not count as beating the census. Beating the census and not STRAW is H5, not senescence. Baker late-life extraovarian is not relabelled an open ovarian window; Farr CTx P = 0.611 and ITP from 20 months are not relabelled a closed ovarian window.
Lineage: L10-4-HG-HT1 v1; L10-4-AR-C4; L10-4-SCOPE-C6; Weenen PMID 14742691; Farr PMID 38956196; Baker PMID 22048312 / 26840489; Harrison PMID 38041783; L10-1-K10.
Kill of the rule: H5 or biomarker is marked, or AMH/Farr/ITP/Baker is used as an ovarian effect.
Kill of the empirical (when an object exists): the STRAW-stratified ovarian Q0 absolute beats out of sample stage + primordial census + systemic.
Status: proposed, parked for human execution.
Maturity: H0. HUMAN_QA_REQUIRED.
Confidence: 0.72 in the rule (AMH out of the census arm); 0.08 in the complete empirical form.
13. Falsifiable predictions and kill criteria
| Hypothesis | Decisive prediction | Result that kills it |
|---|---|---|
AR-H1 | Whole-ovary HMGB1 integer per female does not rise to a threshold written before function; nested 12-section is not another lineage; ABT pool↑ ≠ Flaws P30–60 | The integer rises in two replicates and splits function at matched PMF; or Q0 passes in the primordial as acquired arrest |
AR-H2(2a) | Same-series disector dissolves 951 vs 772, or nuclear FOXO3 / growing ratio align “more pool” with dormancy | Pool↑ is conserved with pinned FOXO3 and Q0 death of a neighbouring mitotic |
AR-H2(2b) | Q0 fails in the non-luteal CDKN2A+ minority at 2 and 9 months, without new exclusions, floor 20 nuclei | Q0 passes and that class’s absolute rises (that tests a threshold; it does not open flavonol) |
AR-H2(2c) | Fibrosis 2 vs 9 months moves with the disector census, not with a Q0 state | Natural fibrosis threshold splits function at matched occupancy, without toxic |
AR-HT1 (rule) | Organ mediation with AMH, AFC, menopause, T-cell p16, Farr, ITP or Baker is not accepted | H5/biomarker is marked or one of those substitutes is used as an ovarian effect |
SCOPE-H1 | Remains H0 on dictyate | A mitotic Q0 (not nested, not cycling epithelium) writes a threshold and splits function |
SCOPE-H2 | D+Q/fisetin do not split Q0 death plus function in the natural | They do so in two replicates at documented occupancy: then flavonol H2 dies and D0-TIME-SPLIT opens |
| Healthspan / G8 | Closed | An organ event with ovarian mediation and competing risks; not AMH nor menopause |
14. Discriminating experiment
This is research. It is not a regimen for a woman. These quantities are not copied as a recommendation. This stage does not gavage. The order is not reopened.
Step 1 — D0-ABS-EXTRACT: zero animals
Only the integer Yan already computed: senescent oocytes/section × total sections on every fifth 10 µm whole-ovary section, plus the primordial census of that series, at 2, 6, 9, 12 and 18 months, per female. Source: source data, supplement, authors or individual points of Fig. 1A and 1F readable as n = 5. Product per female, not product of means. Bars without points = not estimable. Lamin B1, IL-1α, IL-6 and SA-β-gal are archived as a 12-section fraction: they are not extracted as whole-ovary integers (L10-4-ED-C2). GSE253194 does not contain them. This stage did not execute the extraction.
Step 2 — D0-ABS-RECOUNT: same series, only if step 1 is not estimable
C57BL/6J females, no gonadotoxic. Ages 2 vs 9 months written beforehand. Not 18 months (47 ± 14 primordials are a depletion floor, not a ceiling t2). Unit: the female. One ovary to Myers physical disector (primordial class + HMGB1-translocated integer on adjacent sections of that ovary). The contralateral to explant (D0-HMGB1-LOCALE) or archive: never as a class × stain multiplier (L10-4-ED-C3). n = 7 per age. If the HMGB1 integer inherits CV ≈ 0.30 (that of primordials 498/1687 and 72/284), a +50% rise has two-tailed power α = 0.05 ≈ 0.64 at n = 5 and ≈ 0.82 at n = 7 (df = 12, critical t = 2.179). A non-significant 9 vs 2 is non-estimable for a rise, not a ceiling refutation (L10-4-ED-C7). Function coprimary: follicles per female plus corpora lutea of the current cycle. No mating. PBE IVM is not that second readout. Serum AMH, if drawn, is labelled growing pool (Weenen) and does not enter the predicate. Myers’ 1930 ± 286 SEM at 70 days does not calibrate C57BL/6J.
Step 3 — D0-BCL2-CLOCK in the same adults
Flaws P0–P60 is not rerun. ABT is not gavaged to create the sign. Nuclear vs cytoplasm vs mixed FOXO3 (threshold written beforehand; mixed counted separately) and growing:primordial ratio, same female. John’s background is FVB/n: the assay is shuttling, not a transported cut-off. If the authors release Fig. 2 blocks, that series’ disector is applied to that material (the only 6-month object). If not, (2a) is voted in the natural 2 vs 9 months and does not fabricate 951 vs 772 (L10-4-ED-C4).
Step 4 — D0-Q0-MITOTIC in the 2- and 9-month females
EdU 7 days + Ki67− + orthogonal (lamin B1 or focal γH2AX), failed in luteum, atresia and cycling epithelium, without post hoc exclusions. Nested 12-section oocyte out. Sato CL/CA = negative control. Floor ≥20 non-luteal p16INK4a+ nuclei/female. (2b) dies if Q0 fails at both ages (L10-4-ED-C5).
Step 5 — D0-HMGB1-LOCALE in the contralateral explant
Extracellular or surface HMGB1 vs dictyate cytoplasm; TLR4 on granulosa/immune; IL-6 of those neighbours, with the number of cytoplasmic-HMGB1 primaries from the other ovary’s series as a female covariate. A 24 h explant that releases HMGB1 at all ages is not estimable (section injury). Not a biomarker.
Closed from this design
D0-TIME-SPLIT, G1-AGENT, empirical HT1, G8, flavonol or ABT gavage, Flaws P0–P60, 18 months as t2, GSE253194 as EXTRACT. Genetic clearance of a qualified somatic lineage would be preferable to reopening flavonols or navitoclax if G1 died; this stage does not open it. No dose is written.
15. Biomarkers and stratification
There is no validated biomarker. The following are research readouts, not biomarkers:
- Per-female integer of primordial MSY2+ HMGB1-translocated over whole ovary or same-series disector: object of
AR-H1(i), not a clinical threshold. - Nuclear vs cytoplasm vs mixed FOXO3 in adult primordial: dormancy clock, not senotype.
- Durable arrest plus orthogonal that fails in luteum, atresia and cycling epithelium: identity gate (
Q0-STATE). - Fibrosis/collagen correlated with the disector census, not with HMGB1 fraction: clause (2c).
- STRAW+10: minimum human stratification, not a criterion of a trial this stage does not authorize.
- Histological primordial census or dormant nuclear-FOXO3: comparator of
AR-HT1if a Q0 object ever exists. AMH/AFC do not occupy that arm.
p16/p21, SA-β-gal, lipofuscin, CD68, cytoplasmic HMGB1, nested IL-6, AMH, follicles, menstrual regularity, age at menopause, T-lymphocyte p16 and Farr CTx cannot be presented as a single biomarker of ovarian senescent load, health or longevity.
16. Individual variability
Expected heterogeneity includes reproductive stage, basal reserve, reproductive history (breeders vs virgins: Maruyama), toxic and time since injury, immune/CD90+ composition (Isola), luteal state, strain (C57BL/6J vs ICR vs FVB/n), counting method (section density vs disector), PK regimen, and —in humans— STRAW+10, not age alone. Human studies must distinguish:
- natural transition versus iatrogenic ovarian insufficiency or PCOS;
- ovary with follicles versus late postmenopause;
- local action versus systemic disease;
- organ-specific response versus a global index;
- sex: Fang already showed dimorphism under D+Q; Baker 2011 is BubR1 background; Jo (L10-3) is male ICR.
Participation of women is not equivalent to estimating modification by stage (L9-3-K9). Farr’s sixty postmenopausal women without ovary do not estimate STRAW −2. Krishnakumar’s three volunteers do not estimate ovarian occupancy.
17. Pharma relevance and maturity
Pharma relevance is negative derisking, not readiness (L10-4-ED-C6). There is no nominated ovarian senescent-ceiling target. There is no modality. There is no indication in women’s health. There is no STRAW stratum that this stage authorizes. Maturity does not reach H5. HUMAN_QA_REQUIRED before any partnering conversation.
- The title’s object is not a ready-to-evaluate ovarian senolytic calendar. D+Q and fisetin remain H0 of function in García. ABT-263 is a systemic BH3 t1 whose published sign (more primordials when the HMGB1 fraction falls) is incompatible with “I cleared a ceiling” and is not the Flaws census. Navitoclax is not relabelled ovarian senolysis. Venetoclax has approved hematology use; that existence is not a development signal in ovarian aging and this report does not evaluate that indication.
- BCL2 is dual-use. A programme that lowers BCL2 “to clear senescent cells” in ovary and another that raises it “to preserve reserve” cannot share the same ceiling. The laboratory authorizes neither as a recipe.
- Dictyate cytoplasmic HMGB1 is not Davalos’ alarmin. Moving a blocking HMGB1 antibody or a TLR4 antagonist to the ovary because Yan stained cytoplasm copies the category error into an alarmin pipeline.
- AMH is not the primordial census (Weenen). Yan measured AMH and saw it rise with ABT in parallel with the growing pool; that does not convert AMH into human primordial endowment. A development that uses AMH, AFC, menopause, T-lymphocyte p16 or Farr CTx as a surrogate of “ovarian senescent load” violates
AR-HT1. - What would have to exist before a human evaluation, in this order: raw n of the whole-ovary HMGB1 integer or same-series disector that votes parental direction; Q0 in a mitotic, not in prophase I; an absolute of that class that rises before an independent function; matched occupancy of the same intervention; mediation to an organ event with competing risks, beating STRAW + histological primordial census + systemic, not AMH. All of that is H0. Expanding Farr, the ITP, TROFFi or STOP-Sepsis does not fill G7.
Maturity:
- Load threshold on dictyate (
SCOPE-H1): H0. - Primordial non-ceiling (
AR-H1): H1 of literature (category); H0 for “no lineage.” - Rivals 2a/2b/2c (
AR-H2): H0–H1. - Flavonols as an ovarian therapeutic to date (
SCOPE-H2): H1 of literature for García’s quiet function; H0 as proof that no future therapeutic will exist. - Toxic clock (
SCOPE-H3): H1 of literature for t_pre; FOXO3 molecule. - Event translational hypothesis (
AR-HT1): H0, HUMAN_QA_REQUIRED. - Healthspan / G8: closed.
H5 is not marked. Nothing is prescribed.
18. Limitations
- A systematic review with a PRISMA protocol was not performed; the search was directed at the window/load estimand.
- Yan’s raw-n series was not read. A report is not an integer. The product ~134 → ~44 → ~13 remains inferred and, for lamin/IL/β-gal, additionally cropped to 12 sections.
- Yan n = 5; Sato IHC and RNA-seq n = 3; SASP qPCR n = 3. The unit is sometimes the section.
- Ávila 2024 remains at abstract layer; n and dose were not invented.
- Watson is a preprint. It does not fill G7.
- Farr: the map’s 62–88 age range was not reconfirmed in the abstract; it is not used.
- Myers delivers SEM at 70 and 100 days in wild-type of strain undeclared in the abstract. The 1930 does not calibrate the recount.
- Power 0.82 at n = 7 assumes CV = 0.30 inherited from primordials. If the HMGB1 integer is noisier, n = 7 does not reach the +50% rise. Critical t 2.179 is not an ovarian result.
- John is FVB/n, small n, 5 µm formalin; Yan is C57BL/6J, 10 µm frozen. Without Fig. 2 blocks, (2a) does not kill the published sign.
- Davalos is fibroblast. An explant ELISA can measure section injury.
- Saul 2025 does not include ovary. Ki67− does not transport the skin/lung senotype.
- García HTML was not reread this turn; function nulls are inherited from L10-2/L10-3.
p>0.05does not establish equivalence; several “nulls” may be imprecise.- Animal data do not authorize dose or intervention in people.
- The bridge to longevity has no measured event, mediation or competing risks.
- PubMed returned 403 when reconfirming PMID 38871781 this turn; Yan’s figures are held against PMC11176158 already audited in verification, synthesis and design.
19. Conclusions
“Therapeutic window before senescent load is irreversible” names a ceiling that ovarian literature does not deliver. What it delivers is a germ cell already arrested whose fraction of a mislocalized alarmin rises while the parental absolute falls; a systemic BH3 t1 that increases primordials when that fraction falls, on a clock that is not the perinatal Bcl-2 census; two flavonol calendars with quiet function and unequal occupancy; a FOXO3 t_pre that is not senolysis; a p16 maximum in luteum; and a human gap at the first identity link.
The hypotheses that can be falsified now are not a senolytic calendar for women. They are AR-H1 (the whole-ovary HMGB1 integer is not a ceiling), AR-H2 (the residue is dormancy/counting, a mitotic Q0, or natural fibrosis —voted separately, nested out), and AR-HT1 (organ mediation with AMH, Farr, ITP or Baker is not accepted). The smallest discriminant can cost zero animals if Yan releases the series already computed. The first animal, if opened, asks parental direction at 2 versus 9 months in the same series, not senolysis at 18 months on 47 primordials. Women’s longevity is not at stake in any of those endpoints.
